La formiate-tétrahydrofolate ligase est une ligase qui catalyse la réaction :

ATP + HCOO− + tétrahydrofolate    ADP + Pi + 10-formyltétrahydrofolate.

Cette enzyme intervient notamment dans la branche du méthyle de la voie de Wood-Ljungdahl.
